  • Protein tepelného šoku 90 (Hsp90) je molekulární chaperon hojně se vyskytující v eukaryotických buňkách. Jeho přesná úloha však ještě nebyla zcela vyjasněna. Pomocí testu na vazbu železa a analýzy hmotnostní spektrometrií jsme v preparátech membránových proteinů z lidských buněk HeLa identifikovali lidský Hsp90 jako protein vázající železo. Analýza Westernova přenosu a konfokální mikroskopie potvrdila, že část buněčného Hsp90 je spojena s plasmatickou membránou, ale není zřejmě exprimován na buněčném povrchu. Test na vazbu železa s purifikovaným lidským Hsp90 potvrdil, že Hsp90 váže železo. Vyvozujeme z toho, že Hsp90 je protein vázající železo spojený s plasmatickou membránou (cs)

Employing an iron-binding assay and mass spectrometric analysis, we have identified human Hsp90 as an iron-binding protein in membrane protein preparations of human HeLa cells. Western blot analysis and confocal microscopy confirmed that a portion of cellular Hsp90 is associated with the plasma membrane, but it does not seem to be expressed on the cell surface. The iron-binding assay with purified human Hsp90 confirmed iron binding by Hsp90. Thus we suggest that Hsp90 is an iron-binding protein ssociated with the plasma membrane
