| - Měřením inkorporace bromodeoxyuridinu do replikující se DNA jsme zjistili, že imunosupresivní frakce kančí semenné plazmy (ISF) potlačuje proliferaci lymfocytů stimulovanou mitogeny: předběžné působení monoklonální protilátkou proti ISF či indomethacinem účinek ISF zvrátilo. Kromě toho jsme sledovali schopnost indomethacinu či monoklonální protilátky proti ISF zvrátit potlačení produkce protilátek vyvolané ISF měřením titru protilátek v krevním séru myší imunizovaných %22keyhole limpet%22 hemocyaninem pomocí ELISA. V nedávné době jsme popsali ISF jako komplex hlavních semenných glykoproteinů PSP I a PSP II. PSP II je složka odpovědná za imunosupresivní schopnosti tohoto komplexu. Abychom zjistili, zda je imunosupresivní účinek ISF spojen s jeho proteinovou či sacharidovou částí, zkoumali jsme u deglykosylovaného PSP II jeho antiproliferativní účinek na myší lymfocyty stimulované mitogeny. - By measuring bromodeoxyuridine incorporation into replicated DNA, we found that immunosuppressive fraction of boar seminal plasma (ISF) inhibited mitogen-stimulated lymphocyte proliferation; pretreatment with monoclonal antibody to ISF or indomethacin reversed the ISF effect. Besides, we followed the ability of indomethacin or monoclonal antibody to ISF to reverse ISF-induced suppression of antibody production by measuring antibody titers in the blood sera of mice immunized with keyhole limpet hemocyanin using ELISA. Recently we have identified ISF as a complex of the major seminal glycoproteins PSP I and PSP II. PSP II is the part that is responsible for immunosuppressive properties of the complex. To learn whether the ISF immunosuppressive effect is associated with its protein or saccharide part, we examined the deglycosylated PSP II for its antiproliferative effect on mitogen-stimulated mouse lymphocytes.
