Description
| - V dané práci jsme odhalili, že kasein kináza 1 (CK1)delta/epsilon je nezbytná pro fosforylaci proteinu Dishevelled po přidání proteinů z nekanonické Wnt rodiny. Po přidání Wnt-řa, CK1epsilon váže Dv1 a následně ho fosforyluje. Naše data tak poprvé identifikují CK1 jako komponentu signální dráhy indukované proteinem Wnt-5a, která indukuje diferenciace dopaminergních neuronů. Navíc naše výsledky naznačují, že fosforylace Dvl kinázou CK1delta/epsilon je společným prvkem jak v kanonické, tak nekanonické Wnt dráze. (cs)
- Using in vitro gain and loss of function approaches we reveal that casein kinase 1 (CK1)delta/epsilon is critical for dishevelled (Dv1) phosphorylation by non-canonical Wnts. We show that in response to Wnt-řa, CK1epsilon binds Dv1, which is subsequently phosphorylated. Our data identify CK1 as a component of Wnt-řa-induced signalling machinery that regulates dopaminergic differentiation, and suggests that CK1delta/epsilon-mediated phosphorylation of Dvl is a common step in both canonical and non-canonical Wnt signalling.
