  • Efekt exogenní aplikace cytokininu meta-topolinu [mT; N6-(meta-hydroxybenzyl)adenin] v procesu umělé senescence na oddělených listech pšenice (Triticum aestivum L. cv. Hereward) byl studován a srovnáván v listech kde probíhala senescence při kontinuálním světle (100 µmol fotonů m 2 s 1) a ve tmě. Senescencí indukovaná degradace struktury a funkce fotosyntetického aparátu byla charakterizována redukcí obsahu chlorofylu, maximální účinností fotochemie fotosystému (PS) II (Fv/Fm) rychlostí asimilace CO2, hladinou peroxidace lipidů a modifikacemi ve struktuře chloroplastů.Zatímco v oddělených listech uchovávaných ve tmě mT efektivně zpomaloval změny vyvolané senescencí, v segmentech uchovávaných na světle se jeho účinek po několika dnech měnil v opačný. Zde jsme pozorovali přílišnou excitaci PSII, kumulaci škrobových zrn v chloroplastech a stimulaci peroxidace lipidů. Je diskutován pravděpodobný mechanismus akcelerace senescenci-doprovázejícího snížení fotosyntetické funkce a zvýšení peroxidace lipidů ... (cs)

Hereward) was studied and compared in leaves senescing under continuous light (100 µmol photons m 2 s 1) and darkness. Senescence-induced deterioration in structure and function of the photosynthetic apparatus was characterized by reduction in chlorophyll content, maximal efficiency of photosystem (PS) II photochemistry (Fv/Fm) and the rate of CO2 assimilation, by increase in the excitation pressure on PSII (1 - P) and a level of lipid peroxidation and by modifications in chloroplast ultrastructure. While in darkened leaf segments mT effectively slowed senescence-induced changes in all measured parameters, in light-senescing segments the effect of mT changed into opposite a few days after detachment.
