Description
| - Lipidomic analysis is challenging due to enormous complexity of lipid structures. The mass spectrometry (MS) plays a key role in the quantitative lipidomics in separation coupling with MS (mainly LC/MS and SFC/MS). The goal of this work is the development of complementary nontargeted and targeted MS based method for the comprehensive characterization of biological samples, which is applied for the characterization of changes caused by the tumor growth in comparison with surrounding healthy tissues. Comprehensive lipidomic analyses of tumor tissues and surrounding normal tissues from several clinical trials (breast, kidney and lung cancer) were performed using optimized HILIC-HPLC/ESI-MS and SFC/MS. Detailed analysis of lipid species inside above mentioned classes was performed using relative abundances of deprotonated molecules in the negative-ion ESI mode or protonated molecules in the positive-ion ESI mode followed by MS/MS experiments. The statistically significant lipidomic differences were described for different types of tumor tissues (e.g., breast, kidney, lung) in comparison with surrounding normal tissues of the same patient obtained after the surgery.
