| - Jaterní progenitorové buňky mohou být zahrnuty v ranných fázích hepatokarcinogeneze indukované chemickými karcinogeny. Použili jsme potkaní jaterní buněčnou linii WB-F344 ke studiu genotoxických efektů vybraných karcinogenních PAHs. S výjimkou dibenzo[a,l]pyrenu a také částečně benzo[g]chrysenu a benz[a]anthracenu, ostatní studované PAHs indukovaly vysoké hladiny mRNA CYP1A1 a CYP1B1. Pozorovány byly rozdílné genotoxické a cytotoxické schopnosti PAHs. Dibenzo[a,l]pyren a v menší míře benzo[a]pyren, benzo[g]chrysen a dibenzo[a,e]pyren vytvářely vysoké hladiny DNA aduktů, doprovázených akumulací fosforylovaného proteinu p53 a indukcí apoptózy. Naproti tomu benz[a]anthracen, chrysen, benzo[b]fluoranthen a dibenzo[a,h]anthracen indukovaly malé množství DNA aduktů a minimální apoptózu. Jaterní progenitorové buňky by mohly být náchylné k tomuto typu genotoxického poškození, což činí buněčnou linii WB-F344 vhodným nástrojem pro studie genotoxických účinků organických kontaminantů v jaterních buňkách. - Liver progenitor cells may participate at the early stages of hepatocarcinogenesis induced by chemical carcinogens. We employed rat liver WB-F344 cell line to study genotoxic effects of selected carcinogenic PAHs. With exception of dibenzo[a,l]pyrene, and partly also benzo[g]chrysene and benz[a]anthracene, all other tested PAHs induced high levels of CYP1A1 and CYP1B1 mRNA. We observed distinct genotoxic and cytotoxic potencies of PAHs. Dibenzo[a,l]pyrene, and to a lesser extent also benzo[a]pyrene, benzo[g]chrysene and dibenzo[a,e]pyrene, formed high levels of DNA adducts, accompanied with accumulation of phosphorylated p53 protein and induction of apoptosis. Contrary to that, benz[a]anthracene, chrysene, benzo[b]fluoranthene and dibenzo[a,h]anthracene induced only low amounts of DNA adducts formation and minimal apoptosis. The susceptibility of liver progenitor cells to this type of genotoxic insult makes WB-F344 cell line a useful tool for studies organic contaminants effects in liver cells.
