  • Organismy jsou reprezentovány různými variantami zásobních a enzymatických bílkovin. Tento jev bílkovinného polymorfismu je předpokladem využití bílkovin jako genetických markerů, kterých lze využít k identifikaci odrůd ječmene. K získání elektroforetických spekter standardních vzorků semen odrůd jarního a ozimého ječmene registrovaných v ČR byly použity a vzájemně porovnány čtyři postupy elektroforetické separace hordeinových a enzymatických bílkovin ječmene a sice postup elektroforézy ve škrobovém gelu (SGE) podle, postup doporučený mezinárodní organizací semenářské kontroly ISTA, tj. elektroforéza v polyakrylamidovém gelu (PAGE), která je zakotvená v ČSN 46 1085-2, dále metoda elektroforézy v polyakrylamidovém gelu v prostředí SDS (SDS-PAGE), doporučená mezinárodní organizací pro ochranu odrůd (UPOV,1994), alely esterázových lokusů Est 1, Est 2, Est 4 a Est 5 byly stanoveny v plotně polyakrylamidového gelu s tris-glycin pufrem při pH 8,3 ( Sýkorová,1996). The phenomenon of protein polymorphism enables the utilization of proteins as genetic markers that can be used for the identification of the barley varieties. Four procedures for the electrophoretic separation of hordein coming from standard seed samples of barley varieties registered in the CR were used and compared, namely a) starch gel electrophoresis (SGE) according to the Czech State Standard; b) polyacrylamide gel electrophoresis recommended by the International Seed Testing Association (PAGE ISTA); c) SDS-PAGE for hordeins recommended by the Union for the Protection of New Varieties of Plants; and d) PAGE for the determination of alleles of esterase loci Est 1, Est 2, Est 4, Est.5.
