Description
| - The main goal of the project is employment of metagenomic approaches (molecular biology and analytical techniques, analysis of 16SrDNA, ribosomal proteins, functional genes for catabolic enzymes, production of toxins) for characterization of key factors, determining taxonomical and functional diversity of microbial populations dominating in the environment and stressed by different conditions. The profit of such approach is based on possibility to characterize culturable but also non-culturable microbes and characterize their abilities and function on level of DNA (RNA). We suppose to find linkages, interactions and relations critical for indigenous microbial populations in stressed soil systems as a consequence of the effect of different biotic and abiotic factors (presence of allochtonous populations, addition of unusual substrates, presence of plants including transgenic ones, etc.). | - V rámci projektu byla optimalizována metoda analýzy pyrosekvenačních dat, která je vhodná pro studium taxonomické i metabolické diversity bakterií v životním prostředí. S využitím této metodiky spolu s analýzou obsahu kontaminantů v zemině bylo zjištěno, že biostimulací vhodné indigenní mikroflóry lze dosáhnout zrychlení odbourávání PCB z kontaminované zeminy. (cs)
- Methods for analyzing pyrosequencing data have been optimized in the frame of the project. These are suitable for the studies of taxonomic and metabolic diversity of environmental bacteria. Using this methodology along with contaminant concentration assays we found out that biostimulation of certain indigenous microbiota can result in increased degradation rates of PCBs. (en)
