Description
| - The exocyst, a multimeric exocytotic vesicle tethering complex, was extensively studied in yeast and mammals in relation to cell polarity. Recently we confirmed the existence of the whole exocyst complex in plants. Arabidopsis exocyst mutants exhibit defective cell polarity and growth. Cell polarization is a crucial factor for the efficient plant defense on the site of the pathogen attack as cell wall reinforcements (papilla) are deposited through the localized action of the secretory pathway. Our preliminary data show that the formation of the papilla upon a challenge by a pathogen depends on the exocyst function. We propose to explore possible functions of exocyst in plant defense studying Arabidopsis exocyst mutants, especially of the Exo70 family upon pathogen attack (using e.g. Blumeria graminis). The local cell and tissues reactions and localization of different exocyst subunits-GFP constructs will be monitored during the interaction with pathogen. We expect that this project will substantially contribute to the understanding of the role of the secretory pathway in plant resistance to pathogens. (en)
- Exocyst, multimerní komplex účastnící se ukotvování exocytotických váčků je důležitý pro udržování buňečné polarity a dosud byl studován na kvasinkových a živočišných modelech. Nedavno jsme zjistili přítomnost tohoto komplexu i u rostlin. Rostliny Arabidopsis nesoucí mutace v podjednotkách exocystu mají porušenou buňečnou polaritu a růst. Buňečná polarita je klíčovým faktorem pro úspěšnou obranu rostliny proti útoku patogenů, neboť lokalizovaná aktivita sekretorické dráhy je potřebná k zesilování buňečné stěny na místě napadení (ke tvorbě obranné papily). Naše předběžné vysledky nasvědčuji, že tvorba papil je závislá na fungování exocystu. Proto chceme zjistit, jakou roli hraje v rostlinách komplex exocyst při obraně proti útoku patogenů (např. Blumeria graminis) zvl. s využitím mutantů Arabidopsis v podjednotkách exocystu Exo70. Budou sledovány místně specifické buňečné a tkáňové reakce a lokalizace podjednotek exocystu (ve fůzi s GFP) během interakce rostliny s patogenem. Předpokládáme, že tento projekt významně přispěje k pochopení významu sekretorické dráhy pro rezistenci rostlin vůči útoku patogenů.

| - Project solution produced original findings on the role of exocyst in plant immunity and description of some proteins involved. Results were published in 2 papers in top journals in the field (J. Exp. Bot., Plant Cell). While the former is dominantly authored by the project team, including the main and corresponding author, the latter includes only PI and co-PI inside a numerous author team. - Řešení projektu přineslo původní zjištění úlohy exocystu v rostlinné imunitě a popis některých zúčastněných bílkovin. Výsledky byly publikovány ve dvou článcích ve vysoce prestižních časopisech oboru (J. Exp. Bot., Plant Cell), přičemž na prvním dominuje řešitelský tým projektu, včetně prvního a koresp. autora, zatímco na druhém jsou řešitelka a spoluřešitel jedinými členy početného aut. týmu. (cs)
