Description
  • The precise and detailed description of bacterial type strains deposited in microorganisms collections is the basic condition for the control of their identity and authenticity. The type strains are necessary for nearly all taxonomic studies because theyrepresent hitherto described species. Molecular methods based on the analysis of the cellular DNA play an important role together with classical phenotypic tests. Ribotyping represents a molecular method used often in the bacterial taxonomy studies. Thismethod is based on the DNA polymorphism analysis of the genes for ribosomal RNA and the close chromosomal regions. The aim of the project proposed is ribotyping of the type strains of gram-positive cocci and gram-negative rods deposited in the CzechCollection of Microorganisms (CCM). The hybridization profiles database obtained will be a basis for the identification of atypical strains and for the verification of the strain authenticity during a repeated cultivation or the control of strain (en)
  • Přesná a podrobná charakteristika typových kultur bakterií uložených ve sbírkách mikroorganismů je základním předpokladem pro kontrolu jejich totožnosti a autentičnosti. Typové kultury jsou nezbytné pro téměř všechny taxonomické studie jako srovnávacímateriál reprezentující doposud popsané taxony. Mimo klasické fenotypové charakteristiky zaujímají velmi důležité místo i metody založené na analýze buněčné DNA. Jednou v taxonomii bakterií velmi často používanou metodou je ribotypizace, která jezaložena nadetekci DNA polymorfismu v genech pro ribozomální RNA a v jim blízkých regionech bakteriálního chromozomu. Cílem předkládaného projektu je ribotypizace typových kultur gram-pozitivních koků a gram-negativních tyček uložených v České sbírcemikroorganismů (CCM). Vytvořená databáze hybridizačních profilů bude sloužit jako základ pro identifikaci atypických kmenů a pro srovnání a ověření autentičnosti v případě pasážování jednotlivých typových kultur či pro ověření totožnosti typového kmene
