Description
| - Significant inter-species differences have been already clarified in terms of sperm morphology, seminal plasma characteristics, and mechanism of sperm motility. This project deals with a comparative study on metabolic pathways underlying the processes of energy production and cell defense during maturation and ageing of fish spermatozoa between carp (Cyprinus carpio) and sterlet (Acipenser ruthenus). The spermatozoa of these species are considerably different in terms of duration of sperm motility and sperm structure. The main aim is to investigate metabolic pathway in pre-mature, mature and aged spermatozoa by means of cellular and molecular approaches (biochemical assay of bioenergetic pathways and proteomic analysis). In this regard, factors involved in maintaining spermatozoa in quiescent status with adequate level of nutrients for sperm metabolism and protection of sperm against oxidative attacks will be considered. The results will provide us with basic information about mechanisms of sperm maturation and ageing in fish. (en)
- U spermií ryb byly prokázané významné mezidruhové rozdíly v morfologii spermií, v charakteristikách semenné plazmy a mechanismu motility spermií. Tento projekt je zaměřen na komparativní studii metabolických pochodů u spermií kapra obecného (Cyprinus carpio) a jesetera malého (Acipenser ruthenus), které jsou zásadní pro produkci energie a ochranu spermie během dozrávání a „stárnutí“. Spermie uvedených druhů se podstatně liší v pohyblivosti spermií a struktuře spermie. Cílem je studovat metabolické pochody u „nedozrálých“, „dozrálých“ a „stárnoucích“ spermií prostřednictvím buněčných a molekulárních přístupů (biochemické analýzy bioenergetických pochodů a proteomické analýzy). V tomto ohledu budou sledovány faktory podílející se na udržení spermií v „klidovém stavu“ s příslušnou úrovní živin pro metabolismus a faktory podílející se na ochraně spermií před oxidativními změnami. Výsledky nám zprostředkují základní informace o mechanismech dozrávání a stárnutí spermií ryb.
