Description
  • Fatty acids(FA) are recently recognized to be key physiological regulators of oxidative phosphorylation (OxPho), causing partial uncoupling while being translocated via ADP/ATP car- rier as anions and returning protonated across the bilayer, thus resetting efficiency of OxPho. The process is undoubtedly important for cardiology, nutrition and all energetic related physiology. Recently, we have found that FA inhibit and bind to phosphate carrier (PiC), another key point of OxPho. We aim to locate FA binding site in the sequence of PiC by photoaffinity labeling with 3H-azido-FA , enzymatic cleavage and sequencing of resulted peptides and confirm it by site-directed mutagenesis of the given locus. Functionally, it will be investigated, whether FA interfere with a Pi binding site, OH- (or H+) translocation, whether affect HgCl2-induced uniport mode, if FA cycling mechanism is also valid for PiC, etc. (en)
