Description
| - Neuroblastoma, a tumor of the peripheral sympathetic nervous system, is the most frequent solid extra cranial tumor in children and is a major cause of death from neoplasia in infancy. Still little improvement in therapeutic options has been made, requiring a need for the development of new therapies. The project addresses still unsettled questions, which of mechanisms of action of DNA-damaging drugs both currently used for treatment of human neuroblastomas (doxorubicin, cis-platin and etoposide) and another anticancer agent decreasing growth of neuroblastomas in vitro, ellipticine, are predominant mechanism(s) responsible for their antitumor action in vitro (neuroblastoma cells) and in vivo (neuroblastoma xenograft tumours). The project will also investigate the effects of hypoxia on efficiencies and mechanisms of actions of these drugs and whether they are capable of inducing chemoresistance in neuroblastomas. Their effects in combination with histone deacetylase inhibitors and mechanisms of such effects in vitro and in vivo are other subjects, which will be studied. Such a study will increase our knowledge to explain the proper function of these drugs on the molecular level, which should be utilized for the development of new therapies for neuroblastomas. (en)
- Neuroblastomy jsou tumory periferního nervového systému patřící mezi obtížně léčitelné tumory dětské populace. Ačkoliv vývoji nových terapií této choroby se věnuje značná pozornost, stále nevede k uspokojivým výsledkům. Cílem projektu je poznání mechanismů působení protinádorových agens na neuroblastomy in vitro (neuroblastomové linie) a in vivo (experimentální modely s lidskými neuroblastomy). Jedná se o protinádorová agens způsobující různá poškození DNA, a to jak běžně užívaná pro léčení (doxorubicin, cis-platina a etoposid), tak i další látku, které snižuje růst neuroblastomových buněčných linií, ellipticinu. Záměrem projektu je rovněž poznání vlivu hypoxie na účinnost těchto léčiv a jejich mechanismus působení, a poznání jejich potenciálu indukovat chemoresistenci v neuroblastomových liniích. Dalším cílem projektu je poznání vlivu těchto léčiv na růst neuroblastomů in vitro a in vivo a mechanismus jejich působení v kombinaci s inhibitory histondeacetylas. Výsledky získané touto studii vysvětlí aktuální mechanismy působení studovaných léčiv na molekulární úrovni a přispějí k návrhu vývoje nových terapeutických postupů v léčbě neuroblastomů.

| - Hlavním přínosem byly nové poznatky o poškození DNA vybranými cytostatiky na modelu lidských neuroblastomů. Deset stanovených cílů bylo průběžně plněno. Vysoký počet (68) prací s dedikací k projektu v časopisech s nízkým a středním impaktem a rozsáhlá prezentace výsledků špatnou angličtinou v závěrečné zprávě nasvědčují, že projektu by prospěla preference kvality nad kvantitou. - New findings about the DNA damage by selected cytostatics represent the main accomplishment of the project. Ten stated goals were achieved in time. High number of papers (68) dedicated to the project in journals with low or medium impact and extensive presentation of results in final report in imperfect English indicate that the project would benefit from preference for quality over quantity.
