  • 1/ Regulation of mechanism by which corepressors compete with coactivators for binding to androgen receptor. 2/ Elucidate an effect of the inhibitor histone deacetylation on changes of covalent histone modifications. 3/ Determine whether histone modifications changes (acetylation and methylation) stimulate corepressors or they are results of their activation. 4/ If we suppose that methylation could be a reason of androgen receptor silencing, we would like to test the effect of demethylation agents or a its combination with inhibitor of histone deacetylation, on possible expression of androgen receptor.   • 1/ Regulace mechanismu, při kterém se korepresory společně s koaktivátory ucházejí o vazbu s androgenovým receptorem. 2/ Objasnit vliv inhibitoru histonových deacetylaz na změny kovalentních histonových modifikací. 3/ Zjistit, zda-li změny histonových modifikací (acetylace a metylace) stimulují korepresory anebo jsou důsledkem jejich aktivace. 4/ Předpokládáme-li, že metylace může být příčinou umlčení exprese androgenového receptoru, ověřit účinek demetylačního agens případně v jeho kombinaci s inhibitorem histonových deacetyláz na obnovenou expresi androgenového receptoru.
