Description	Analysis of germline p53, CHK2 and SNF5 gene mutations, rearrangements and polymorphisms in families with hereditary breast cancer and other malignancies where no BRCA1/2 mutations were found will be performed. Molecular genetic analysis of tumours will also be performed,e.g.loss of heterozygosity and p53 protein stability. It is likely that further candidate genes will be published, and these will also be studied. Information materials for medical professionals and patients will be published. Analysis of data from our database will concentrate on genotype-phenotype correlations and putative anticipation in affected families. The project will contribute to lowering of fraction of families with a striking hereditary predisposition but with no genetic cause identified. The project results will have impact on counselling and care for affected individuals. (en) Bude provedena analýza zárodečných mutací, přestaveb a polymorfismů v genech p53, CHK2 a SNF5 v rodinách s dědičnou rakovinou prsu a dalšími malignitami, u kterých nebyly nalezeny mutace v genech BRCA1/2. Molekulárně geneticky budou analyzovány též nádory, zejména ztráta heterozygozity a stabilita proteinu p53. Je pravděpodobné, že budou publikovány další kandidátní geny, a ty budou také analyzovány. Budou vypracovány informační materiály pro lékaře a postižené. Analýza dat soustředěných v naší databázi se pokusíme odhalit korelace mezi genotypem a fenotypem a možnou anticipaci v postižených rodinách. Projekt přispěje ke snížení podílu rodin se zřetelnou dědičnou predispozicí ale bez odhalené genetické příčiny. Výsledky projektu budou mít důsledky pro poradenství a péči o postižené.
