Description	WT1 gene is potentially an important marker with hematological malignancies. Its role in malignant and normal cells is presumably influenced by the expression pattern of its isoforms and by the mutation state. In the proposed study, using original methodology we will identify expression patterns of WT1 isoforms of childhood AML, ALL, MDS. We will correlate the results with the mutation state of the gene and other biological and clinical properties of the individual disease subtypes. We will compare the expression patterns of malignant cells with the subgroups of physiological progenitors in bone marrow. We will thus answer some controversial questions, which cast doubts on the role of the WT1 gene as a marker for malignant transformation, disease progression and monitoring MRD and immunotherapy for pediatric malignancies. We will establish a new differentially diagnostic procedure using the quantification of the WT1 expression in combination with polychromatic flow cytometry with childhod MDS. (en) Gen WT1 je potenciálně důležitým markerem hematologických malignit. Jeho roli v maligních i normálních buňkách pravděpodobně ovlivňuje expresní vzorec jeho isoforem a mutační stav. V navržené studii určíme pomocí originální metodiky kvantitativně expresní vzorce isoforem WT1 u dětských AML, ALL a MDS. Budeme výsledky korelovat s mutačním stavem genu a dalšími biologickými a klinickými vlastnostmi jednotlivých subtypů onemocnění. Porovnáme expresní vzorce maligních buněk i s podskupinami fysiologických progenitorů v kostní dřeni. Zodpovíme tak některé kontroversní otázky, zpochybňující roli exprese genu WT1 jako markeru maligní transformace, progrese nemoci a cíle pro sledování MRN a imunoterapii u dětských hematologických malignit. Dále stanovíme nový diferenciálně diagnostický postup využívající kvantifikaci exprese WT1 v kombinaci s polychromatickou průtokovou cytometrií u dětských MDS. (cs)
