Description	Cephalosporins are indispensable antibiotics for therapy of severe infections in humans and animals, resistance to these antibiotics has increased. The project suggests to characterize a collection of coliform bacteria resistant to cephalosporins from livestock, humans, the environment and populations of wild animals. The aim is to determine genetic basis of cephalosporin resistance and characterize mobile genetic elements carrying these genes including the possibilities of transfer to susceptible strains. Molecular analysis of plasmids from strains from different environments and geografical areas and their comparing will help to undestand the circulation of these elements and the role of their co-selection by antibiotic therapy. Obtained data will be used for the risk assessment of their dissemination and for modification of antibiotic policy in Czech Republic. The results will contribute to understand the epidemiological connection between dissemination of specific plasmids carrying ESBL genes in Europe and will help to determine their evolution in various geographical areas. (en) Cefalosporiny jsou nepostradatelnými antibiotiky pro léčbu závažných infekcí člověka i zvířat, rezistence bakterií k těmto antibotikům nárůstá. Projekt navrhuje charakterizovat soubor izolátů koliformních bakterií rezistentních k cefalosporinům z hospodářských zvířat, lidské populace, prostředí a populací volně žijících zvířat. Cílem projektu je determinace genetického základu rezistence k cefalosporinům a charakterizace mobilních genetických elementů nesoucích tyto geny včetně možnosti jejich přenosu na kmeny citlivé. Molekulárně-biologická analýza plazmidů z kmenů z různých prostředí a geografických oblastí a jejich srovnání umožní objasnit cirkulaci těchto elementů a úlohu jejich koselekce antibiotickou terapií. Získané výsledky budou podkladem pro stanovení rizik jejich šíření a pro případnou úpravu antibiotické politiky v ČR. Získané výsledky přispějí k odhalení epidemiologických souvislostí v rozšíření specifických plazmidů s geny pro ESBL v Evropě a napomůže stanovit zákonitosti jejich evoluce v různých geografických oblastech.
