Description
  • Kallikrein 14, a newly identified serine protease of the kallikrein family, is thought to be involved in the function of skin homeostasis, i.e. migration, differentiation and desquamation of keratinocytes. To elucidate the physiological role of KLK14 in vivo, that shows exceptional activity in the epidermis, we plan to generate conditional knockout mice with targeted deletion of the Klk14 gene. To analyze the function of Klk14 in pathological processes transgenic mice with conditional overexpression of Klk14 in the skin should be generated using a lentiviral vector, which enables a time-controlled transgene expression.The impact of Klk14 deficiency and overexpression in the skin will then be studied in different models of skin inflammation targeting the epidermal barrier function and the skin immune system. Characterization of the role of kallikrein 14 may have significant clinical implications for the treatment of skin disorders in which KLK activity is dysregulated. (en)
